Periodized Mitochondrial Stack Strategy

One of the most important aspects of building an effective mitochondrial-based stack is choosing a primary anchor compound—a central agent that the rest of your stack revolves around. This is the compound I like to build everything around, adjusting the supporting elements based on the phase of the protocol.

Over time, I’ve found the best results by rotating through three core mitochondrial compounds, each with a specific role in the cycle. This periodization helps avoid adaptation and maximizes mitochondrial efficiency and cellular energy output.

Here’s how I structure my 16-week mitochondrial optimization cycle:


Weeks 1–3: SS-31 (Priming Phase)

  • Dose: 1–3mg per day

  • Purpose:

    • SS-31 is the mitochondrial primer.

    • It helps repair and stabilize mitochondrial membranes, improve ATP production, and reduce oxidative stress.

    • This phase is critical for preparing the mitochondria to respond more effectively to the rest of the stack.


Weeks 4–8: MOTS-C + NAD+

  • Dose:

    • MOTS-C: 5mg, 3x per week

    • NAD+ (injectable or liposomal): according to personal tolerance (typically 100–300mg per dose if injectable)

  • Purpose:

    • This stack enhances metabolic flexibility, insulin sensitivity, and AMPK activation.

    • MOTS-C and NAD+ synergize well to support mitochondrial biogenesis, cellular resilience, and energy production.

    • Best used together during this active rebuilding phase.


Weeks 8–12: 5-Amino-1MQ (Solo Phase)

  • Dose: 1–3mg per day

  • Purpose:

    • 5-Amino-1MQ supports mitochondrial health by inhibiting NNMT, increasing NAD+ retention, and promoting muscle performance and fat loss.

    • Personally, I find this compound works best as a solo agent rather than stacked with MOTS-C.

    • This phase often delivers a noticeable “spark” in energy output and performance.


Weeks 13–16: Fasting + Anti-Senescent Protocol

  • Purpose:

    • This is a recovery phase focused on cellular cleanup and reset.

    • Incorporate intermittent or prolonged fasting, along with senolytic agents (like FOXO4-DRI or fisetin) to help the body clear senescent cells and prepare for another cycle.

    • This break helps maintain sensitivity to mitochondrial enhancers and avoid diminishing returns.


Core Add-Ons (Used Throughout All 12 Weeks)

Regardless of the main anchor, I always keep these foundational mitochondrial enhancers in play:

  • Methylene Blue – Low-dose daily for mitochondrial electron cycling and cognitive support

  • Ubiquinol – A bioavailable form of CoQ10 for ATP production

  • PQQ – Supports mitochondrial biogenesis and antioxidant defense

  • SLU-PP-332 – Boosts energy expenditure and mitochondrial activity


Why I Rotate Instead of Running One Compound Long-Term

Staying on a single compound for the full 12 weeks often leads to plateaus. By rotating between SS-31, MOTS-C + NAD+, and 5-Amino-1MQ, I’ve noticed:

  • Better adaptation and cellular responsiveness

  • Enhanced performance consistency

  • Less biological “dullness” compared to single-compound monotony

Each agent offers unique mitochondrial mechanisms, and rotating them allows for a layered, phased approach to optimization—priming, building, then amplifying.
