Tropisetron Protocol 

Tropisetron is a unique and highly underrated nootropic. Mechanistically, it works by blocking the 5-HT3 serotonin receptors in both the gut and brain—this is why it’s often associated with reduced nausea. In addition, it acts as a partial agonist at the α7-nicotinic acetylcholine receptor, which is one of the more interesting pathways tied to cognition, inflammation control, and dopamine modulation.

Dosing Approach

Most people begin at 5 mg per day and increase slowly based on response.
For me personally, 10 mg was the “sweet spot.” Some researchers go as high as 20 mg, but in my opinion:

I wouldn’t use it continuously for more than 2–3 weeks due to the limited long-term human data available.

My Personal Experience

While using Tropisetron, I noticed:

  • Significantly less brain fog

  • Reduced inflammation

  • A clear dopamine uplift, likely tied to 5-HT3 receptor blockade

  • More stable mood and better cognitive flow
